while I'm rightclick on the file which in the project,the upload option is blank,can't select,is there something must be setup first?

I suspect that your file must be part of a project, and the project must have an account (e.g. FTP) assigned to it.

I had already created a FTP account,in the menu of tools-->account,but,how to assigned it to an project?!

Use the Project Properties dialogue box (on the Tools menu), and assign the FTP account in the Publishing section. If you have not created a project yet, do New Project on the File menu
